Organophosphate Pesticide Exposure Reduced Serum Paraoxonase1 (PON1) Activity Which Correlated With Oxidative Stress in Pesticide Factory Workers
Authors
Abstract:
Background & Aims of the Study: Serum paraoxonase (PON1) is a potent antioxidant that is associated with the pathogenesis of several diseases. Also, it is reported that environmental factors can modulate the PON1 activity. In this study, the association between the Organophosphates (OP) exposure and plasma Paraoxonase/Arylesterase (PON1) activity and also OP-induced oxidative stress was investigated among pesticide manufacturing factory workers. Materials and Methods: In this cross-sectional study, Plasma cholinesterase (PChE) and RBC Acetylcholinesterase activity (AChE) were measured as a biomarker of OP exposure. Also, the amount of malondialdehyde (MDA) was evaluated as a biomarker for lipid peroxidation and oxidative stress. The PON1activity was measured by two distinct substrates, Paraoxson and Phenylacetate, to measure paraoxonase (PONase) and arylesterase (AREase) activities of PON1 enzyme respectively. Results: A significant decrease was observed inPChE and AChE activities in the blood sample of factory workers which confirmed OP exposure (p<0.001). Furthermore, the level of MDA increased that shown lipid peroxidation induced by OP (p<0.05). PONase and AREase activities decreased significantly in factory workers (p<0.05, p<0.001_). The reduction in AREase activity shown a correlation with the decrease in PChE and AChE activities (p<0.01), also a negative correlation was observed with AREase activity and lipid peroxidation (p<0.01). The Ponase activity only negatively correlated with lipid peroxidation (p<0.01). Conclusion: Our study indicates that occupational OP pesticide exposure in factory workers causes a decrease in serum PON1 activity which is associated with oxidative stress. Decreased PON1 activity is involved in the pathogenesis of several diseases such as cardiovascular disease then it is recommended that PON1 activity of OP-pesticide workers has been periodically monitored.

The Changes of Erythrocyte Indices in Pesticide Factory Workers
Abstract Background and Objectives: Erythrocyte indices are helpful for monitoring the erythrocyte synthesis and liver condition. The pesticide factories’ staff are being exposed to poisoning. This article aims to evaluate the effect of pesticides on blood indices of subjects which are exposed to organophosphate toxins. Material and Methods: This cohort study was conducted on the 63 emplo...

Prenatal organophosphate (OP) pesticide exposure has been reported to be associated with adverse birth outcomes and neurodevelopment. However, the mechanisms of toxicity of OP pesticides on human fetal development have not yet been elucidated. Our pilot study birth cohort, the Study of Asian Women and Offspring's Development and Environmental Exposures (SAWASDEE cohort) aimed to evaluate enviro...

INTRODUCTION Organophosphate (OP) pesticides remain widely used in agriculture. Previously, we reported that PON1 genotype was directly associated with neurodevelopment at age two, and that PON1 genotype may increase susceptibility to OP exposure. OBJECTIVES We examined the relationships of maternal and child PON1 genotype and enzyme activity levels and neurodevelopment at school age and exam...
full textSerum Cholinesterase Inhibition in Relation to Paraoxonase-1 (PON1) Status among Organophosphate-Exposed Agricultural Pesticide Handlers
BACKGROUND Animal studies have demonstrated that low paraoxonase-1 (PON1) status (i.e., low catalytic efficiency and/or low plasma PON1 activity) is associated with neurotoxic effects after exposure to several organophosphate (OP) insecticides. However, few human studies have investigated associations between PON1 status and intermediate end points, such as serum cholinesterase [butyrylcholines...

INTRODUCTION Acute exposure to pesticide due to suicidal poisoning is the most extensive cause of pesticide exposure, compared with all other causes including agricultural or industrial exposure. Organophosphate (OP) and carbamate group of pesticides can inhibit acetylcholinesterase; on the other hand, paraoxonase1 can detoxify organophosphate poisoning by hydrolyzing organophosphate metabolite...

Organophosphate (OP) pesticides are commonly used in the United States, and farmworkers are at risk for chronic exposure. Using a sample of 218 farmworkers in 24 communities and labor camps in eastern Washington State, we examined the association between agricultural crop and OP pesticide metabolite concentrations in urine samples of adult farmworkers and their children and OP pesticide residue...
